HGH Fragment 176-191 (AOD-9604) is a modified form of amino acids 176 through 191 from the C-terminus of human growth hormone. The full somatropin molecule has separate domains responsible for different aspects of its activity — the N-terminal region is primarily responsible for anabolic and growth-promoting effects via IGF-1 stimulation, while the C-terminal region around residues 176-191 is the area believed to mediate the lipolytic (fat-breaking) activity. The fragment was developed specifically to isolate that lipolytic activity.

What the research covers

Animal studies with AOD-9604 showed reductions in body fat, particularly visceral and subcutaneous, without the IGF-1 elevation, glucose dysregulation or tissue growth effects seen with full somatropin. The mechanism appears to involve beta-3 adrenergic receptor stimulation and direct effects on fat cell metabolism. It entered human clinical trials for obesity but did not demonstrate sufficient efficacy to proceed to Phase 3. The research interest remains in the principle of isolating the lipolytic activity of GH from its anabolic effects — an approach that has informed subsequent fat-loss peptide research.
