NAD+ (nicotinamide adenine dinucleotide) is a coenzyme present in every cell in the body. It sits at the centre of cellular energy metabolism — it is the electron carrier that shuttles reducing equivalents through the mitochondrial electron transport chain to produce ATP. It is also the substrate consumed by sirtuins (SIRT1–7) and PARP enzymes, which govern DNA repair, gene regulation and stress response. NAD+ levels decline measurably with age and in conditions including obesity and neurodegeneration, which is the research rationale for supplementation.

What the research covers

The most studied mechanisms involve sirtuin activation and PARP-mediated DNA repair. Animal studies using NAD+ precursors (NMN, NR) have demonstrated improvements in muscle function, metabolic health and lifespan in multiple model organisms. Direct NAD+ infusion work in humans has examined cognitive function, energy metabolism and fatigue in small pilot studies. David Sinclair’s group at Harvard has been the most prominent in positioning NAD+ biology as a central longevity pathway. The intravenous route bypasses the gut conversion steps required by oral precursors, which is the research rationale for the injectable format.

Where the evidence sits

Human trial data on injectable NAD+ specifically is limited — most published work uses oral precursors. Animal longevity data is consistent but has not translated cleanly to human trials yet. NAD+ is not licensed as a medicine. Supplied strictly as a laboratory research chemical.
